In the preferences pane you are just changing where Zotero looks for its directory, not the location of the directory itself. So you need to find the Zotero folder and move it to its new location.

You might try this approach. Form the export pane of the preferences window select BibTeX from the drop down menu. Then use the Ctrl+Shift+C (Command+Shift+C on a Mac) keyboard shortcut to copy any selected items to your clipboard.

Sadly this is a limitation of RIS, I do not believe they support a field for call number. "N1" is the notes field in RIS, so it looks like Endnote is dumping call numbers into notes because there is no field for it.
