Vancouver style (as described here: http://www.ncbi.nlm.nih.gov/books/NBK7256/ ) explicitly doesn't call for spaces there, if you do, you're not using Vancouver. It's easy enough to change that in a style and there are numbered styles that do have a space after the comma.
