After automatic update, clicking Zotero in Safari has no response.

I have already tried restarting the browser, uninstalling and reinstalling the Zotero Connector, updating the translator, and other troubleshooting steps, but the Zotero extension still does not respond at all when clicked in Safari.

Agree with whoislazymouse, and the link provided does not fix or answer the issue.

I don't know what "agree with" is supposed to mean. The linked page explains that this is a longstanding macOS bug and provides the only known ways to fix it. If you follow the steps carefully, one of them will almost always work, but if you're still having trouble, then as the page says, you should use a browser with a more reliable extension framework.
  • What it means is that following the steps outlined on the page does not work, or no difference is apparent.

However, I just quit Safari, reopened it, and now it works. There is a line on the page that offers a note ("It may also help to restart your computer in between deleting the app and restoring it, though this isn’t usually necessary."), but no mention of quitting Safari and restarting the browser.

Because it's not usually necessary, but restarting your computer would obviously also restart Safari.
